You need to do BOTH.
First, turn off the printer. This will effectively clear the printer's 
memory, BUT it will not remove the print job from your computer.
Next, use the lprm command (or CUPS) to cancel the print job.

The lp commands are:
lpq #this will tell you what jobs are queued up. If you have more than one 
printer, use the -P (upper case P) option.

lprm # Remove a job from the queue. You get the job number from the lpq 
command. Again, the -P option can be used to point at the specific printer.

These will work with CUPS. Additionally, printer queues survive reboots. 

Once all the jobs are removed, then it is safe to turn on your printer. The 
memory should be cleared.
